Becker & Hickl DCS-120 MP Multiphoton FLIM Confocal System

This unique bespoke system brings together a wide range of cutting-edge techniques that are not commonly available, all within a single platform based on a Zeiss Axio Observer 7 inverted microscope. Its core innovation is the combination of tunable pulsed excitation from a femtosecond laser source (Insight X3, Spectra-Physics), an acousto-optic modulator and a second harmonic generation module (APE HarmoniXX Select SHG) with single photon counting detectors and time-correlated single photon counting (TCSPC) electronics from Becker & Hickl.

The counting electronics have an exceptionally fast time response (<40 ps, compared to 100 ps on the previous FLIM system), allowing quantitative determination of fluorescence observables with unprecedented accuracy. This makes the system a powerful and versatile platform for advanced lifetime, anisotropy and correlation-based imaging.

  • Excitation: femtosecond pulsed laser (Insight X3) with AOM and SHG module (HarmoniXX Select)

  • Detectors: 2 x single photon counting detectors (HPM-100-07) on the confocal and non-descanned channels

  • TCSPC electronics: SPC-180NX, time response <40 ps

  • Objectives: 4x, 20x, 40x and 63x oil

  • Fluorescence lifetime imaging (FLIM) with both 1P and 2P excitation (340–650 nm and 680–1300 nm)

  • Time-resolved fluorescence anisotropy imaging (trFAIM)

  • Phosphorescence lifetime imaging (PLIM)

  • Fluorescence correlation spectroscopy (FCS)

  • Second harmonic imaging (SHIM)

  • Non-descanned detection for deep-tissue multiphoton imaging
